目的探讨核因子κB(NF-κB)诱捕脱氧寡核苷酸(ODNs)对子宫内膜息肉(EP)间质细胞在白细胞介素1β(IL-1β)诱导培养下环氧化酶2(COX-2)合成及雌激素(E2)分泌的影响。方法 15例EP患者行间质细胞的分离培养,分为3组:刺激(A)组,用10μg/L的IL-1β刺激培养;转染后刺激(B)组,NF-κB诱捕ODNs转染EP间质细胞后,用10μg/LIL-1β刺激培养;空白对照(C)组。培养12、24、36和48h,RT-PCR检测细胞中COX-2水平,ELISA检测细胞培养上清中E2水平。结果 A组自12h开始COX-2合成显著高于C组,自24h开始E2的分泌量显著高于C组(P〈0.01)。B组COX-2合成及E2分泌较A组显著下降(P〈0.01)。结论 NF-κB诱捕ODNs能显著抑制IL-1β诱导的EP间质细胞合成COX-2及分泌E2。
Objective To explore the effect of neuclear factor(NF)-κB decoy ODNs on the synthesis of cyclooxegenase(COX)-2 and estradiol(E2) in stromal cells of endometrial polyps(EP) induced by interleukin(IL)-1β.Methods Stromal cells from 15 EP cases were purified,cultured and divided into 3 groups with 5 cases each.Group A were cultured with 10 μg/L of IL-1β,group B was transfected by NF-κB decoy ODNs and then cultured with 10 μg/L of IL-1β.Group C was taken as blank control.At 12,24,36 and 48 h,the expression of COX-2 in stromal cells was detected by RT-PCR and the concentration of E2 in the supernatant was measured by ELISA.Results The expression of COX-2 since 12 h and E2 secretion since 24 h were significantly higher in group A than those in group C(P0.01),which were significantly lower in group B than those in group A(P0.01).Conclusion NF-κB decoy ODNs can significantly decrease the COX-2 expression and E2 secretion stimulated by IL-1β in stromal cells of EP.
