目的建立一种简便、有效的小鼠滋养层细胞原代培养方法,为研究病理妊娠的发生机制奠定基础。方法分别在K.Handschuh和ZhouWH方法的基础上进行改良,获取孕鼠滋养层细胞,进行原代培养,并应用细胞计数及免疫细胞化学法比较滋养层细胞数量和纯度。结果K.Handschuh改良法和ZhouWH改良法获得滋养层细胞数目分别为(11.4±1.8)×10^4个+/mL、(16.3±0.9)×10^4个/mL,纯度分别为95%、96%,在获得滋养层细胞数目方面ZhouWH改良法优于K.Handschuh改良法(P〈0.05)。结论Zhou WH改良法可获得高纯度足量的小鼠滋养层细胞。
Objective To explore an optimized method of primary culture of murine trophoblast cells so as to lay foundation of study on mechanism of pathogenic pregnancy. Methods Trophoblast cells were derived from pregnant miee through modified methods of Handschuh and Zhou WH and cultured primarily. The number of trophoblast cells was counted by cell counter, and their purity was tested by immunoeytochemistry. Results The number of trophoblast cells derived through modified Handschuh method was ( 11.4 ± 1.8) ×10^4/mL, and that through modified Zhou WH method was ( 16.3 ±0.9)×10^4/mL. Their purities were 95% and 96% respectively. In derived number of trophoblast cells, the modified Zhou WH method was superior to the modified K. Handschuh method. Conclusion Highly pure and large number of trophoblast cells can be derived through modified Zhou WH method.
