以N-羟基琥珀酰亚胺(NHS)和1-(3-二甲氨基丙基)-3-乙基碳二亚胺盐酸盐(EDC·HCl)为催化剂,将制备的双羧基聚乙二醇(COOH-PEGCOOH)和部分巯基化聚乙烯亚胺(PEI-SHx )先后接枝于壳聚糖分子,得到巯基化壳聚糖衍生物(CS-PEGPEI).利用傅立叶红外光谱仪(FT-IR)和核磁共振仪(1H NMR)对其结构进行表征;通过透析法制备自聚集纳米粒,透射电子显微镜(TEM)和动态激光粒度分析仪(DLLS)分析测试自聚集纳米粒显示,自聚集纳米粒为球状纳米胶束,该纳米球有望成为纳米药物及基因的载体.
Thiolated chitosan derivative (CS-PEG-PEI) was synthesized by grafting double carboxy-terminated PEG and partly thiolated polyethylenimine (PEI-SHx) onto chitosan with NHS/EDC as catalyst. The chemical structure of polymer was characterized by the Fourier transform infrared spectroscopy(FT-IR) and proton nu- clear magnetic resonance(1 H NMR), the self-assembled nanoparticles was prepared by probe sonication-dialysis method. The measurements of transmission electron microscope(TEM) and dynamic light scattering(DLS) in- dicated that the self-aggregates were nanoparticles. This novel nanoparticle system was promising to be a good drug carrier and gene carrier.
