Objective To compare the multi-generational reproductive and developmental outcomes in rats which consume 3 types of drinking water with different mineral levels. Methods Ninety female and forty-five male weaning SD rats were randomly divided into 3 groups, with the ratio of female to male of 2 to l per group. The 3 groups were given 3 different waters separately from the weaning rat till to whose F3 generation pups weaning. The animals were allowed free access to food and water. The 3 types of water were water A [ total dissolved solids (TDS), 1.2 mg/L], water B (TDS 87.2 rag/L) and C (TDS 291.0 mg/L). Parental reproductive outcomes and offspring developmental parameters were observed and compared in each generation. Results There were no statically differences in the parental reproductive parameters in all the 3 groups from FO to F2 generations. The offspring survival rates (OFSR) on postnatal days (PND) 4 in F3 generation were statistically lower in water A group than the other 2 groups (P 〈 O. 01 ). While the OFSR on PND4 of the water C group was not only obviously higher in F1 generation than those of the water B group (P 〈0.01), but also notably higher in F3 generation than those of the other 2 groups. What's more, the learning ability of the pups in water C group was higher than the other 2 groups. Conclusion Drinking water which hardly contains minerals might have no effect on the parental reproductive ability, but have negative effect on the early development of the offspring. For the rats in early period of development, drinking water with hygienic safety and rich minerals is beneficial to the development.
