目的探讨加巴喷丁对关节炎大鼠背根神经节成纤维细胞生长因子2(fibroblast growth factor-2,FGF-2)及成纤维细胞生长因子受体1(fibroblast growth factor receptor-1,FGFR1)表达的影响。方法成年健康雄性SD大鼠30只,随机分为3组:对照组、模型组和加巴喷丁组。模型组和加巴喷丁组大鼠注射弗氏完全佐剂诱导关节炎模型。加巴喷丁组在建模后第7~14天腹腔注射加巴喷丁。检测大鼠造模后的第1、4、7、10、14天足底厚度和缩足反射机械刺激阈值(paw withdrawal mechanical threshold,PWMT)的变化情况;免疫组织化学法检测造模后第7天及第14天大鼠背根神经节内FGF-2及FGFR1的表达。结果与正常组大鼠相比,模型组及加巴喷丁组大鼠出现关节炎症状,PWMT值明显下降(P〈0.05),背根神经节内FGF-2及FGFR1表达增多,加巴喷丁组给药后PWMT升高,FGF-2及FGFR1表达量下降。结论加巴喷丁能减轻关节炎性痛并降低背根神经节FGF-2及FGFR1的表达。
This study designed to explore the effect of gabapentin on the expression of FGF-2 and FGFR1 in dorsal root ganglion neurons of arthritis rats. Adult healthy male SD rats were randomly divided into 3 groups (n= 30): control group, model group and gabapentin group. The model and gabapentin group rats were injected with complete Freud adjuvant to induce arthritis. The gahapentin group rats were injected with gabapentin once a day from the 7th day to 14th day after adjuvant injection. The plantar thickness and paw withdrawal mechanical threshold (PWMT) of rats were record at the 1st, 4th, 7th, 10th, 14th day; the expressions of FGF-2 and FGFR1 were observed at the 7th and 14th day. Compared with control group rats, the model and gabapentin group rats showed the symptoms of arthritis, decreased PWMT (P〈0.05), and increased expression of FGF-2 and FGFR1 in the dorsal root ganglion. After gabapentin injection, the PWMT was increased, FGF-2 and FGFR1 expressions were decreased. This study suggests that gabapentin can relieve the suffering of arthritis and decrease the expression of FGF-2 and FGFR1 in dorsalroot ganglia.
