目的 探讨经腹腔注射锰后,大鼠大脑核受体相关因子1(Nurr1)的改变. 方法 健康雄性2月龄SD大鼠50只,按体重随机分为5组:生理盐水组(A)、Mn2+ 2.5 mg/kg(B)、Mn2+5 mg/kg(C)、Mn2+ 10 mg/kg(D)、Mn2+ 20mg/kg (E),每天腹腔注射0.5ml锰溶液,连续染毒30 d.实时荧光定量PCR检测大鼠脑组织中Nurr1 mRNA的相对表达水平,Western blot检测Nurr1的蛋白表达水平. 结果 脑组织Nurr1 mRNA相对表达的结果显示,与A组相比,B、C、D组Nurr1 mRNA含量降低,分别是A组的0.85,0.66,0.54和0.33倍,经多组比较的单因素方差分析显示不同浓度组间差异有统计学意义(P<0.01).同时,脑组织Nurr1蛋白含量也随染锰剂量增大而降低,经多组比较的单因素方差分析显示差异有统计学意义(P<0.01).结论 锰可以在mRNA和蛋白质水平改变Nurr1的表达.
Objective To explore the changes of Nurr1 expression in brain after exposure to manganese (Mn2 +) in rats.Methods Fifty male SD rats were randomized into 5 groups:normal saline group,2.5 mg/kg Mn2+ group,5 mg/kg Mn2+ group,10 mg/kg Mn2+ group,20 mg/kg Mn2+ group.They were exposed to Mn2+ by intraperitioneal injection at the dose of 0.5 ml/d for 30 d.The mRNA expression of Nurr1 in brains in rats was measured by real-time fluorescence quantitative PCR (RT-PCR).The Nurr1 protein levels in brains of rats were measured by Western blot.Results RT-PCR results showed a 0.85,0.66,0.54 and 0.33 fold decrease of Nurr1 mRNA expression in groups B,C,D,and E respectively compared with normal saline group(P < 0.01).Western blot results showed a gradual reduction of Nurr1 protein in groups B,C,D,and E compared with normal saline group(P < 0.01).Conclusion Manganese can alter the levels of Nurr1 in mRNA and protein expression in brain of rats.
