以厚皮甜瓜鲁厚甜2号为材料,在绿色叶肉组织特异表达启动子PNZIP驱动下,利用农杆菌介导的遗传转化系统,将小麦抗真菌γ-硫堇蛋白(γ-Thionin)基因(THI)导入甜瓜,获得卡那霉素抗性植株。转化植株经PCR和Northern blot检测分析,证实γ-硫堇蛋白基因已经转入甜瓜基因组中并在甜瓜叶片组织中表达。
Antifungal γ- thionin gene Till which had been cloned from Triticum aestivum and driven by green specific promoter(PNZIP) was transferred into muskmelon by Agrobacterium tumefaciens. Transgenic melon lines carried witb both γ- THI gene and npt- Ⅱ gene were assessed by PCR and Northern blotting. The resluhs showed that the foreign genes were proved to be transferred into the genome of muskmelon and expressed in leaf tissue of the transgenic plants.
