Due to the frequent movement of some types of protozoan in water, it is a challenging problem to count them. The purpose of this study is to design a new digital image processing technology to solve the problem. Two or more photos of the protozoan were continuously taken by a camera. Then they were transformed into a black and white picture using some image processing measures, such as image subtracting, enhancement, filtering and binarizing. The resulting image was further processed to identify and label connected regions, followed by automatically counting the regions and statistically revising the result. An automatic counting system was developed to count the ciliate, such as paramecium, and obtained more accurate and efficient results as compared to the manual counting method.
