目的 探讨丹酚酸B(SalB)对脑缺血高能磷酸化合物和乳酸含量的影响.方法 复制急性脑缺血模型,测定缺血30 min脑组织高能磷酸化合物和乳酸的含量以及脑组织的含水量.结果 缺血组脑组织ATP[(12.12±3.71) μmol/g]和磷酸肌酸[(65.24±8.84) μmol/g]含量及Na+-K+-ATPase活性[(1.321±0.373)U/mg]显著降低(P<0.01),而乳酸[(3.58±0.81) mmol/g]含量显著升高(P<0.01)、脑指数[(1.508±0.002)%]和脑含水量[(78.75±0.47)%]明显增多(P<0.05~0.01).用丹酚酸B治疗,脑组织ATP[(18.16±3.38) μmol/g]和磷酸肌酸[(74.46±6.58) μmol/g]含量显著升高(P<0.01),Na+-K+-ATPase[(1.902±0.491) U/mg]活性增强(P<0.05),而乳酸[(2.59±0.41) mmol/g]含量明显减少(P<0.01)、脑指数[(1.505±0.002) %]和脑含水量[(78.10±0.44) %]则降低(P<0.05).结论 增加高能磷酸化合物以及降低乳酸的含量,可能是丹酚酸B减轻急性脑缺血早期脑水肿的作用机制.
Objective To investigate the effects of salvianolic acid B (SalB) on the contents of energy rich phosphate and lactic, acid in mice with cerebral ischemia. Methods The model of acute cerebral ischemia was established and the contents of energy rich phosphate and lactic acid, and brain water content were detected 30 minutes after brain ischemic. Results In the ischemic group the cerebral contents of ATP [ ( 12.12 ± 3.71 ) μmol/g ] and creatine phosphate [ (65.24 ±8.84 ) μmol/g ], the activity of Na + - K + - ATPase [ (1. 321 ±0. 373) U/mg] were obviously decreased (P 〈0.01 ), while the content of lactic acid [ (3.58 ± 0.81 ) mmol/g] (P 〈 0.01 ), cerebral index [ ( 1.5(18 ± 0. 002) % ] and brain water content [ ( 78.75 ±0. 47 ) % ] were increased obviously ( P 〈 0.05 0. 01 ). In the SalB treated group the cerebral contents of ATP [ ( 18.16 ±3.38) μmol/g] and creatine phosphate [ (74.46±6.58) μmol/g] were obviously increased (P 〈0.01 ), the activity of Na + -K + - ATPase [ ( 1. 902 ± 0. 491 ) U/mg ] was improved ( P 〈 0.05 ), while the content of lactic acid [ (2. 59 ±0.41 ) mmol/g] (P 〈0. 01 ), cerebral index [ ( 1. 505 s0.002) % ] and brain water content [ (78.10 ±0.44) % ] were decreased (P 〈 0.05 ). Conclusion The mechanism of SalB for relieving early cerebral edema after acute cerebral ischemia may be related to that SalB can increase the content of energy rich phosphate and decrease the content of lactic acid.
