目的观察大鼠肺移植后肺组织高迁移率族蛋白B1(HMGB1)表达和血清HMGB1水平的改变。方法 42只大鼠随机分为对照组(6只)、移植2h组(12只)、移植6h组(12只)和移植12h组(12只),三套管法建立大鼠左肺原位移植模型。使用实时荧光定量逆转录多聚酶链反应检测肺组织HMGB1mRNA水平,使用免疫印迹法和免疫荧光染色分析肺组织HMGB1蛋白水平。血清中HMGB1、TNF-α、IL-1β和IL-6含量采用ELISA法检测。结果大鼠肺移植后2h肺组织HMGB1mRNA表达和HMGB1蛋白含量即见显著升高(P〈0.01),并随时间延长而增强。大鼠肺移植后各时间点血清HMGB1、TNF-α、IL-1β和IL-6水平均明显升高(P〈0.01)。结论大鼠肺移植术后HMGB1表达发生明显改变,HMGB1参与肺移植缺血再灌注损伤过程。
Objective To investigate the changes of HMGB1 expression in lung tissue and HMGB1 level in serum, after rat lung transplantation. Methods 42 rats were randomly divided into control group (n= 6), post-transplantation 2 h group (n= 12),post-transplantation 6 h group (n= 12) and post-transplantation 12 h group(n= 12). The experi- mental orthotopic lung (left) transplantation model in rats was established by cuff technique. The expression of HMGB1 mRNA was quantitatively detected by real-time RT-PCR,and HMGB1 protein levels by Western blot and im- munofluorescence staining,in lung tissue. H MGB1 , TNF-α, IL-1β and IL-6 in serum were analysed using ELISA. Results HMGB1 mRNA and HMGB1 protein expression in lung tissue significantly increased 2 h after lung transplantation (P〈0.01), and were more elevated following post-transplantation hours. The levels of H MGB1, TNF-α, IL-1β, and IL- 6 in serum were markedly increased at each time point after lung transplantation (P〈0.01). Conclusion HMGB1 ex- pression significantly change after lung transplantation in rats,and HMGB1 may participate in lung ischemia/ reperfu- sion injury after lung transplantation.
