目的:观察缺氧对人肺成纤维细胞高迁移率族蛋白Bl(HMGBl)释放的诱导作用。方法:采用HFLl人肺成纤维细胞株.观察缺氧培养不同时间对细胞HMGBlmRNA表达和培养上清液中HMGBI含量的影响。及不同浓度N一乙酰半胱氨酸对HMGBl释放的抑制作用。HMGBl含量采用酶联免疫吸附试验检测。结果:缺氧培养6h后HMGBImRNA表达水平和培养上清液中HMGBl含量明显升高(P〈0.01),后者随着培养时间延长而进一步升高;10mmol/L和100mmol/LN一乙酰半胱氨酸对缺氧培养HMGBl释放有不完全的抑制作用。结论:缺氧能诱导人肺成纤维细胞释放HMGBl,其机制可能与胞内氧自由基产生有关。
Objective To investigate the induction of extracellular release of high mobility group box 1 (HMGB1) in human lung fibroblasts induced by hypoxia. Methods The changes of HMGB1 mRNA expression and HMGB1 concentration in the culture medium were investigated after HFL1 human lung fibroblasts were subjected to hypoxia (1% 02) for different times. Inhibitory effect of N-acetylcysteine on HMGB1 release was observed. HMGB1 concentration was determined by enzyme-linked immunosorbent assay (ELISA). Results The levels of HMGB1 mRNA expression and HMGB1 concentration in the culture medium were significantly increased after human lung fibroblasts were subjected to hypoxia for 6 h (P 〈 l).01), and were increased as the induction time lengthened. 10 mmol/L and 100 mmol/L of N-acetylcysteine can partly inhibit extracellular release of HMGB1 in hypoxia-cuhured human lung fibroblasts. Conclusion Hypoxia induces human lung fibroblasts to release HMGB1, in which mechanism may be involved with intracellular production of reactive oxygen species.
