目的:观察载银二氧化钛(TiO:)纳米管涂层种植体的体内生物相容性,探讨不同表面涂层体内异物反应(Foreign Body Response,FBR)程度。方法:分别对纯钛抛光组(trr)、TiO,纳米管组(NT)、低浓度载银TiO:纳米管组(NAg-L)和高浓度载银TiO:纳米管组(NAg—H)种植体的表面形貌进行扫描电镜观察。将4组种植体(各12枚)植入SD大鼠背部皮下,饲养12d后取材。采用免疫荧光染色计数测量纤维包裹层内CD68阳性的巨噬细胞密度;组织学染色观察纤维包裹层厚度;实时定量PCR分析周围组织巨噬细胞趋化蛋白( Monocyte chemoattractant protein, MCP-1 )的表达量。结果:纤维包裹层内巨噬细胞密度、各组周围组织中MCP-1mRNA表达量均以胛组最多,与其他各组相比均有统计学意义(P〈0.05);而NAg—L、NAg—H、NT3组间无统计学差异(P〉0.05);各组包膜厚度为NT组〈NAg—L组〈NAg—H组〈PT组。结论:低浓度银纳米颗粒载人不增强材料的异物反应,高浓度银轻微增强其异物反应。
AIM: To observe the in vivo biocompatibility of titanium dental implant with surface modified by Agloaded TiO2 nanotube array. METHODS: The implant surface was modified by polished titanium(PT), TiO2 nanotube array( NT), Ag-loaded TiO2 nanotube array with low density( NAg-L ) and Ag-loaded TiO-2 nanotube array with high density(NAg-H) respectively. The surface morphology of the samples in the 4 groups was examined by SEM. Then 12 implants of each group were subcutaneouly transplantated into the back of SD rats. 12 d after transplantation foreign body reaetion(FBR) was assessed by quantification of CD68-positive macrophages in the capsule and by measuring the reactive capsule thickness. The mRNA expression of monocyte chemoattraetant protein-1 ( MCP-1 ) was measured by RT-PCR. RESULTS : The density of macrophages and MCP-1 mRNA expression were significantly high- er in PT group than in other groups (P 〈0.05). However, there was no statistical difference among NAg-L, NAg-H and NT groups( P 〉 0.05 ). Reactive capsule thickness was in the order of NT group 〈 NAg-L group 〈 NAg-H group 〈 PT group. CONCLUSION: Agloaded TiO-2 nanotube array with low density does not raise the foreign body reaction, however high density slightly enhances the foreign body reaction.
