以苎麻雄性不育保持系GS13-X1为实验材料,选用MS培养基附加激素TDZ和2,4-D,研究不同激素浓度组合对种子苗叶片和茎段外植体愈伤诱导与不定芽分化的影响.试验结果表明,GS13-X1再生的最适培养基为MS+0.3 mg/L TDZ+0.05 mg/L 2,4-D+30 g/L蔗糖+7.5 g/L琼脂,叶片和茎段的愈伤诱导率分别为100%和95.24%,出芽分化率分别为11.11%和23.81%.50 mg/L卡那霉素可以明显抑制茎段和叶愈伤诱导与分化.苎麻雄性不育保持系组织培养再生技术是苎麻雄性不育转基因验证的前提条件,也是苎麻遗传工程改良的基础.
Plant regeneration of maintainer line 'GS13-X1' of male sterility in ramie(Boehmeria nivea(L.) Gaud.) was studied.The optimal regeneration medium,MS medium with 0.3 mg/L TDZ,0.05 mg/L 2,4-D,30 g/L sugar and 7.5 g/L agar induced 100% and 95.24% calluses from seedling leaves and stems respectively,and 11.11% and 23.81% shoots from the calluses.50 mg/L kanamycin remarkably inhibited the callus induction and shoot regeneration.The technique for plant regeneration of ramie maintainer line is the foundation for validation of the candidate genes of ramie male sterility.
