细胞凋亡与许多人类疾病的发生密切相关,过度凋亡引发神经退行性疾病,而抑制凋亡则与肿瘤等疾病的产生有关。研究表明,Bcl-2蛋白家族在细胞凋亡中发挥重要作用,特别是近几年对Bcl-2蛋白家族成员Mcl-1及其抑制剂研究越来越多。本文介绍了Mcl-1蛋白的结构与功能特点,综述了近几年作为抗肿瘤药物的Mcl-1抑制剂的研究进展。
Apoptosis is closely related to the occurrence of many human diseases. Excessive apoptosis causes neurodegenerative diseases, while insufficient apoptosis is related to the generation of cancers and other diseases. Nowadays, Bcl-2 family proteins are found to be important in apoptosis regulation and many inhibitors of Bcl-2 have been investigated. Recently, more and more researchers focus on the studies of Mcl-1, which is a member of Bcl-2 family proteins, due to its unique role in the apoptosis pathway. This paper summarizes the structure and function of Mcl-1, and the recent progress of Mcl-1 protein inhibitors as potential antitumor agents.
