目的 初步探讨利培酮和消幻汤合并使用对谷氨酸功能低下小鼠模型表现出的高活动性及偏好行为的影响.方法 昆明种小鼠70只随机分为五组,其中四组腹腔注射溶于生理盐水的地卓西平马来酸盐(MK-801)[(0.072 mg/mL,5 mL/(kg·d)]14 d,第5组作为空白组腹腔注射生理盐水作为相同干预.造模14 d后,利培酮组、消幻汤组、利培酮及消幻汤合并使用组(合并组),灌胃给予相应药物4周;模型组和空白组灌胃给予水作为安慰剂.同时采用自主活动仪、避暗仪、跳台仪分别观察五组的自主活动、偏好行为的变化.结果 模型组在第14天与空白组比较,自主活动次数有显著性增高[(36.8±16.2)次比(11.3±14.5)次,P<0.05)];给药4周后各给药组的高活动性被抑制,各给药组与空白组比较差异无统计学意义(P> 0.05);各给药组的探究活动在造模时期和给药时期差异均无统计学意义(P>0.05).偏好行为结果显示,合并组在给药结束时与造模结束时相比,小鼠的避暗和跳台潜伏期分别延长34.1、20.2 s(P< 0.05).结论 消幻汤和利培酮合并使用能抑制精神分裂症模型小鼠的自主性活动增多,并能延长偏好行为中损害记忆时间,对模型小鼠具有保护作用.
Objective To investigate the effect of Risperidone and Xiao Huan Tang on activity and preferences behavior of glutamate dysfunction mice model.Methods 70 Kunming mice were randomly divided into 5 groups,one group was chosen as blank group.Rest groups were intraperitoneal injected MK-801 [(0.072 mg/inL,5 mL/(kg·d)] continuously for 14 d,then randomly numbered:model group,Risperidone group,Xiao Huan Tang group and Risperidone combined Xiao Huan Tang group.Each group were garaged corresponding drugs for one month,at the same time high activities and changes in the preferences of five groups were observed.Results Compared with the blank group of (11.3±14.5) times,activity of model group of 06.8±16.2)times increased (P < 0.05).After gavaged 4 weeks,model groups of high activity decreased,especially Risperidone combined Xiao Huan Tang group.There was no statistical meaning in inquiry activity of five groups (P > 0.05).Compared with at the end of molding model,latent period of step-through test was prolonged of 34.1 s,of step-down test was prolonged 20.2 s at the end of administration in Risperidone combined Xiao Huan Tang group (P< 0.05).Conclusion The combination of Xiao Huan Tang and Risperidone can suppress the high activity,prolong harmed memory time,and protect preference behavior of schizophrenia mice.
