探讨人低分化食管鳞癌细胞系EC-109、CaEs-17在缺氧条件下,缺氧诱导因子-1α及血管内皮生长因子随缺氧时间不同,在基因水平和蛋白水平上的表达.方法:1)体外培养人低分化食管鳞癌细胞系EC-109、CaEs-17,利用免疫荧光染色方法观察细胞在缺氧培养条件下HIF-1α和VEGF蛋白表达情况.2)提取总RNA,逆转录为cDNA,并进行实时PCR,观察细胞在不同缺氧时间,hif-1α及vegf基因表达峰度及相互关系.结果:1)EC-109、CaEs-17细胞在缺氧培养条件下HIF-1α、VEGF蛋白表达增加.2)EC-109、CaEs-17细胞在缺氧培养条件下hif-1α、vegf基因表达上调.3)EC-109、CaEs-17细胞在缺氧条件下vegf表达峰度上调滞后于hif-1α表达.自缺氧培养8h起,hif-1α基因表达峰度持续高于vegf基因表达.结论:缺氧环境是诱导人食管癌细胞缺氧诱导因子-1α和血管内皮生长因子表达上调的因素之一,vesf基因表达滞后于hif-1α基因表达.
Objective: To investigate the expression of hypoxia inducible factor-1α(HIF-1α) and vascular endothelial growth factor (VEGF) at the mRNA and protein levels in esophageal squamous cell carcinoma (ESCC) cell lines EC-109 and CaEs-17 at different time points under hypoxic conditions. Methods: Human ESCC cell lines EC-109 and CaEs-17 were cultured in vitro and the expression of HIF-1α and VEGF proteins under hypoxic conditions was detected with immunofluorescence. Real-time PCR was used to detect HIF-1α and VEGF mRNA expression. Results: The mRNA and protein expression levels of HIF-1α and VEGF in EC-109 and CaEs-17 cells were increased under hypoxic conditions. The increase in VEGF mRNA and protein expression appeared later than that of HIF-1α under hypoxic conditions, and HIF-1α mRNA and protein expression was higher than VEGF mRNA and protein expression at 8 hours after induction of hypoxia. Conclusion: Hypoxic conditions can upregulate the mRNA and protein expression of HIF-1α and VEGF in ESCC cells. The increase in VEGF expression appears later than that of HIF-1α.
