开口箭对小鼠5-羟色胺分泌的影响

目的:采用灌胃、石蜡切片、免疫组化染色和ELISA等方法,探讨开口箭对小鼠5-羟色胺(5-HT)分泌的影响.方法选取20~22g健康昆明小鼠20只,均分成A,B,c,D4组,分别以蒸馏水和0.25、0.50、1.00mg/mL的开口箭灌胃,0.20mL/次/d,连续灌胃7d.第8d对全部小鼠摘眼球取血,ELISA检测血清5-HT质量浓度;取血后立即处死小鼠,取十二指肠,4%甲醛固定,石蜡切片,免疫组化染色,镜下统计5-HT阳性细胞数量.结果与A组相比,B,C,D组小鼠血液5-HT质量浓度逐渐降低,十二指肠肠壁组织中5-HT阳性细胞数也明显减少.结论开LI箭会下调小鼠5-HT的分泌.

英文摘要:

Objective: By the methods of gavage, paraffin section, immunohistochemical staining, and ELISA, the effects of Tupistra chinensis on 5-hydroxytryptamine secretion was studied in mice. Methods: Twenty healthy Kunming mice weighing 20-22 g were selected and divided into A, B, C and D groups on average. In a serial 7 days, drenched respectively distilled wa ter to A group mice and 0.25, 0.50, 1.00 mg/ml Tupistra chinensis diluted solution to B, C and D group mice, with the dose of 0.2 ml/t/d. At 8th day, the blood of all the mice was collected by the method of eyeball picking and the content of 5- hydroxytryptamine in blood serum was analysis by the method of ELISA, and meanwhile, all of mice were killed rapidly, the duodenums were harvested, fixed in 4 ~//00 formaldehyde, made paraffin sections, stained by immunohistochemical methods, and 5-hydroxytryptamine positive cells were observed and count under light microscope. Results: From A to D group mice, the content of 5-hydroxytryptamine in blood serum were decreased gradually, and also the quantities of 5-hydroxytryptamine posi- tive cells in duodenums were less and less. Conclusion: Tupistra chinensis can downregulate secretion of 5-hydroxytryptamine in mice.
