目的探讨钥孔戚血蓝蛋白(KLH)诱导Balb/c小鼠脾细胞Th1/Th2失衡的特点,为药物干预Th1/Th2失衡奠定基础。方法以KLH+完全弗氏佐剂(CFA)免疫Balb/C小鼠并分离其脾细胞,ELISA法检测脾细胞上清中Th1型细胞因子IFN-γ、IL-2、IL-12p40和Th2型细胞因子IL-4、IL-5,以及血清Th1型抗体IgG2a和Th2型抗体IgG1水平。结果①免疫小鼠出现脾肿大、脾细胞计数升高。②免疫小鼠脾细胞IL-2于24h明显升高,48h达高峰;IL-4、IFN-γ和IL-5于24h轻度升高,96h达高峰;各时间点IL-12p40均较低。③不同剂量基础或强化免疫均可致细胞因子升高,IL-4/IFN-γ升高。④免疫小鼠血清IgG1、IgG2a升高,以IgG1明显。结论KLH+CFA可诱导Balb/c小鼠脾细胞发生Th2型优势反应。
Objective To investigate the characteristics of Th1/Th2 imbalance in splenocytes induced by keyhole limpet hemocyanin (KLH) in Balb/c mice, and to establish the base of medicine interfering in Th1/Th2 imbalance. Methods Female Balb/c mice were immunized with KLH emulsified in complete freund' s adjuvant(CFA), and the splenocyts were separated. The production of Th1 cytokines IFN-γ,IL-2 ,IL-12p40 and Th2 cytokines IL-4,IL-5 in splenocytes culture supernate and specific KLH antibody IgG2a and IgG1 in blood serum were detected by ELISA. Results (1)The spleens derived from KLH immunized mice appeared hypertrophic and the number of splenocytes was manifold. (2)IL-2 production was elevated after incubated for 24 hours, and achieved peak at 48 h; IL-4, IL-5 and IFN-γ was elevated after 24 h, and elevated gradually to 96 h; IL-12 p40 production was very low at every time points. (3) Using different doses of KLH or fortified immunization could all lead to the elevated production of cell factors and lead to the elevation of IL-4/IFN-γ ratio. (4)The levels of serum IgG2a and IgG1 of immunizated mice were elevated,especially IgG1. Conclusions Female Balb/c mice immunized with KLH and CFA can induce Th2 predominance imbalance in splenocytes.
