To find out the optimized conditions for electroporation of Escherichia coli JM109, the optimal electroporation protocol would be found out through changing some important effecting factors, such as the growth phrase of the germ, temperature, voltage, etc. It showed that the optimal conditions were cells harvested in OD600 0. 5--0. 7, capacitance 25 μF, electric resistance 200 Ω, voltage 2. 5 kV, cuvette with an interelectrode distance of 0. 2 cm. Under this condition, the transformation efficiency is 9.12 ×10^8.
