目的研究何首乌提取物对鱼藤酮所致帕金森病线虫模型的神经保护作用。方法设对照组、鱼藤酮组和用药组(0.5 g/L,1.0 g/L),观察其对线虫多巴胺神经元树突、体内活性氧水平及生存率的影响。结果与对照组相比,0.5 g/L和1.0 g/L何首乌提取物对帕金森病模型多巴胺树突减少表型有明显的抑制,显著增加正常多巴胺神经元树突的线虫比例,活性氧水平明显低于鱼藤酮组,线虫存活率明显高于鱼藤酮组。结论何首乌提取物提高线虫生存率,对鱼藤酮致多巴胺树突减少表型有抑制作用,何首乌提取物对鱼藤酮所致帕金森病线虫模型有神经保护作用,其作用机制与抑制活性氧生成有关。
Objective To study the neuroprotective effect of polygonum extract on rotenone- induced Parkinson' s disease model nema-tode. Methods The experienment set control group, rotenone group and treatment group( 0. 5 g/L, 1. 0 g/L), and their effect on nematodes dopamine neurons dendrites, and oxygen levels influence survival in vivo activity were observed. Results Compared with the con-trol group, the 0. 5 g/L, 1. 0 g/L Polygonum extract on Parkinson disease model significantly inhibited dopamine phenotype of dendrit-ic, showed a significant increase in dopamine neuron dendrites normal proportion of nematodes, the reactive oxygen species was significantly lower than the rotenone group, and the nematode survival rate was significantly higher than rotenone group. Conclusion Polygon-um extract improves the nematode survival, inhibits rotenone- induced dopaminergic phenotype of dendritic. Polygonum extract has neuro-protective effects on rotenone- induced Parkinson' s disease, and its mechanism of inhibition is related to active oxygen production.
