目的:探讨汉防己甲素对人胃癌BGC-823细胞株的增殖抑制与诱导凋亡的作用.方法:通过MTT实验检测汉防己甲素对于细胞生长的抑制率,流式细胞术检测细胞凋亡比,Western blotting检测凋亡相关蛋白表达.结果:汉防己甲素显著抑制BGC-823细胞生长,呈时间-剂量依赖性.流式细胞术检测显示不同浓度汉防己甲素干预后,凋亡细胞比例明显增高,且呈浓度-时间依赖性.Western blotting检测显示Bax,Bad和Bak蛋白表达增加,Bcl-2和Bcl-xl蛋白表达降低,Caspase-3和Caspase-9表达增加,用Caspase抑制剂z-VAD-fmk预处理4h后,z-VAD-fmk能抑制汉防己甲素引起的Caspase-3表达增加.结论:在一定条件下,汉防己甲素能够抑制胃癌BGC-823细胞增殖并且诱导其发生凋亡.
Objective:To study the effects of Tetrandrine on proliferation-inhibited and apotosis-induced effect of gastric cancer BGC-823 cells.Methods:The growth of BGC-823 cells treated with Tetrandrine was investigated by MTT proliferation assays.Cell apoptosis was quantified with flow cytometry using Annexin-V-PI staining.The expression of apoptosis-related protein was analyzed by Western blotting.Results:Tetrandrine treatment showed strong growth inhibitory and apoptotic effects on human gastric cancer BGC-823 cells in a time-and concentration -dependent manner.Western blotting showed the up-regulation of Bax,Bak and Bad,down-regulation of Bcl-2 and Bcl-xl,the activation of Caspase-3 and-9.Meanwhile,pretreatment with the pan-Caspase inhibitor z-VAD-fmk reduced Tetrandrine-induced apoptosis by blocking activation of Caspases.Conclusion:In certain conditions,Tetrandrine could inhibit proliferation and induce apoptosis in human gastric cancer BGC-823 cells.
