目的研究罗格列酮对去势和不去势大鼠骨密度和骨微结构的影响。方法取32只6月龄SD雌性大鼠随机分为两组,双侧卵巢摘除或仅切除双侧卵巢周围少量脂肪组织,术后三周将全部大鼠随机分为四组,每组8只。sham(R)组和OVX(R)组以罗格列酮每日2 mg/kg溶解于1 m1生理盐水灌胃,Sham(NS)组和OVX(NS)以生理盐水每日1 ml灌胃8周,观察大鼠股骨和腰椎骨密度、骨微结构等指标。结果去势组腰椎骨密度明显降低,OVX(NS)组和OVX(R)组较sham(NS)组分别降低8.5%和14%,OVX(R)组较sham(R)组降低9.1%。OVx(R)组股骨和腰椎骨小梁骨量较sham(NS)组分别减少41.3%和87.5%,较sham(R)组减少37.9%和78.9%。OVX(R)组股骨和腰椎骨小梁厚度较sham(NS)组分别减少11.8%和41.4%,较sham(R)组减少11.8%和33.3%。结论罗格列酮可使大鼠骨密度降低,骨小梁骨量减少,在雌激素缺乏时,罗格列酮对大鼠骨骼破坏加倍,骨密度显著降低,骨微结构明显受损。
Objective To investigate the effects of rosiglitazone on bone mineral density and bone microstructure in rats.Methods Thirty two SD female rats aged 6 months were ovariectomized or sham operated.After three weeks,they are randomly assigned into four groups(n = 8 in each):the sham(R) group and the OVX(R) group received intragastric administration of rosiglitazone 2 mg for 8 weeks,and the sham(NS) group and the OVX(NS) group received intragastric administration of normal saline(1 ml ? day~(-1)) for 8 weeks.Bone mineral density(BMD) of femur and lumbar were determined by dual-energy X-ray absorptiometry(DXA).Bone microstructure including trabecular bone volume and trabecular thickness were measured by micro-CT.Results Compared with sham (NS) group,the BMD of lumbar of OVX(NS) group and the OVX(R) group declined by 8.5%and 14%,respectively.The OVX(R) group declined by 9.1%compared with group sham(R). Compared with the sham(NS) group,the trabecular volume of femur and lumbar in OVX(R) group decreased by 41.3%and 87.5%;while the OVX(R) group decreased by 37.9%and 78.9% compared with the sham(R) group.In addition,the trabecular thickness of femur and lumbar of rats of the OVX(R) group is lower than the sham(NS) group,by 11.8%and 41.4%,while they declined by 11.8%and 33.3%compared with the sham(R) group.Conclusion Rosiglitazone may decrease the BMD and trabecular volume of rats,particularly for the ovariectomized rats.
