采用适当的料液比、提取温度、提取时间、提取pH对银杏叶进行浸提,采用Sevage法将提取液中的蛋白从粗多糖中去除,经DEAE-52和Sephadex G-100纯化得到均一多糖。以VC为参照,分别研究GBLP对超氧阴离子自由基、羟基自由基和1,1-二苯基-2-苦味酰基自由基的清除作用。结果表明,以料液比1∶40、温度100℃、时间4h、pH 5.0为提取条件时,提取率可达到12.8%。通过测定GBLP对超氧阴离子自由基、羟基自由基和1,1-二苯基-2-苦味酰基自由基的清除率可知,GBLP具有较强的抗氧化性,可作为天然抗氧化剂。
Polysaccharide in Cn'nkgo biloba leaves (GBLP) was prepared and its antioxidation was determined. GBLP was extracted from Ginkgo biloba leaves by water extraction and alcohol precipitation and proteins were removed by Sevage method. The crude polysaccharides were purified by DEAE-52 and Sephadex G-100. The extraction rate of GBLP was 12.8% at the optimal extraction condition of solid-liquid ratio 1 : 40, extraction temperature 100℃, extraction time 4 h, extract pH 5.0. Results suggested that GBLP could be used as a natural antioxidant with high antioxidant activity.
