目的探讨NGF、BDNF是否参与和促进糖尿病视网膜病变新生血管的形成。方法将研究对象分四组:正常健康(CG)组20例,糖尿病无视网膜病组(NDR)50例,非增生性糖尿病视网膜病变(NPDR)组50例,增生性糖尿病变视网膜病变(PDR)50例。收集临床资料:年龄、糖尿病类型、糖尿病病程、收缩压(SBP)、舒张压(DBP)、糖化血红蛋白(HbAlc)、尿素氮(BUN)、肌酐(Cr),采用酶联免疫吸附测定法(ELISA)检测外周血血浆NGF、BDNF含量。结果PDR患者血浆中NGF、BDNF水平(5.014±0.536ng/ml,(2.679±0.637)ng/ml高于CG组(3.549±0.426)ng/ml,(1.135±0.394)ng/ml、NDR组(3.556±0.962)ng/ml,(1.375±0.944)ng/ml、NPDR组(3.732±1.363)ng/ml,(1.504±1.191)ng/ml,比较差异有统计学意义。血浆NGF、BDNF水平与糖尿病病程、HbAlc有正相关性(NGF:r=0.821,0.828;BDNF:r=0.876,0.902,P〈0.01)。结论糖尿病视网膜病变患者的外周血NGF、BDNF水平升高,并与糖尿病病程、HbAlc的水平成正相关。糖尿病视网膜病变患者的外周血NGF、BDNF水平的升高,可能参与促进了视网膜新生血管的形成。
Objective To measure serum nerve growth factor(NGF)and brain-derived neurotrophic factor (BDNF)concentrations in diabetic patients to determine whether NGF and BDNF correlated with angiogenesis in proliferative diabetic retinopathy (PDR).Methods All participators were divided into four groups, 20 nondiabetic controls and 50 non-diabetic retinopathy (NDR)patients and 50 non-proliferative diabetic retinopathy(NPDR)patients and 50 PDR patients.Patient demographic characteristics and diabetic parameters, including age, diabetes duration, HbA1c, renal function, were evaluated in two separate university hospitals.Serum NGF and BDNF concentrations were determined by using the ELISA technique. Results Serum NGF and BDNF levels were found to be higher in PDR patients (5.014±0.536)ng/ml, 2.679± 0.637)ng/ml than in nondiabetic controls (3.549 ± 0.426)ng/ml, ( 1.135 ± 0.394)ng/ml and NDR patients(3.556± 0.962)ng/ml, ( 1.375± 0.944)ng/ml and NPDR patients(3.732± 1.363 )ng/ml, ( 1.504± 1.191 ) ng/ml. NGF and BDNF levels correlated well with diabetes duration, HbA1c (NGF: r = 0.821,0.828 ; BDNF: r=0.876,0.902,P 〈0.01 ).Conclusions Elevated NGF and BDNF levels in PDR patients correlate well with diabetes duration, HbA1c.NGF and BDNF correlate with angiogenesis in PDR.
