目的探讨协同利用卵泡抑素、骨形成蛋白和Wnt11对人骨髓间充质干细胞向心肌细胞分化的影响。方法培养的人骨髓间充质干细胞发育到二代后,分别加入卵泡抑素、骨形成蛋白和Wnt11,用相差显微镜观察细胞的形态变化。收集细胞,RT-PCR检查NKx2.5和MEF2c基因表达的变化。结果卵泡抑素、骨形成蛋白6和Wnt11诱导了人骨髓间充质干细胞出现类似于心肌细胞的形态学变化及心肌细胞形成有关基因的表达。结论协同利用卵泡抑素、骨形成蛋白和Wnt11可以诱导人骨髓间充质干细胞向心肌样细胞方向分化。
Objective To investigate the effect of follistatin, bone morphogenetic protein and Wntl 1 on the differentiation of human mesenchymal stem cells (MSC) into cardiomyocyte. Methods Human bone marrow was collected and mononuclear cells were isolated. MSC from passage 2 were cultured in conditional mediums containing follistatin, bone morphogenetic protein and Wntll at 37℃ in a humid 5 % CO2 air atmosphere. Cardiac differentiation of MSC was assessed at different time points by morphology and with the specific markers of cardiomyocyte. Results The combined use of follistatin, bone morphogenetic protein and Wntl 1 induced MSC to differentiate toward the cardiomyocytes as demonstrated by the morphology observed under phase-contrast photography and expression of some specific genes by RT-PCR. Conclusions The combined use of follistatin, bone morphogenetic protein and Wntl 1 can induce MSC to differentiate towards cardiomyocytes.
