目的研究白头翁皂苷D(毛茛科植物白头翁Pulsatillachinensis(Bge.)Regel的干燥根)体内抗小鼠H22肝癌细和体外的抗人肝癌SMMC-7721作用。方法在体外实验中,采用MTT法考察白头翁皂苷D对SMMC-7721细胞增殖的抑制作用;在体内实验中,采用小鼠H22实体瘤进行体内抑瘤实验,测定肿瘤生长抑制率,利用光镜(HE染色)、电镜等方法进行形态学观察。结果白头翁皂苷D处理人肝癌SMMC-7721细胞24、48、72h后,给药组均出现了不同程度的细胞增殖抑制。白头翁皂苷D(4.5、6.0mg/kg)对小鼠H22肝癌细胞实体瘤的生长有明显的抑制作用,抑瘤率分别为38.67%和41.59%(P〈0.01,与模型组比较)。组织学观察显示,肿瘤细胞大量变性坏死,电镜下可见凋亡细胞和凋亡小体。结论白头翁皂苷D具有一定的体内、外抗肿瘤作用。
AIM To study the effect of Pulsatilla saponin D from root of Pulsatilla chinensis (Bge.) Regel on human hepatocarcinoma cell line SMMC-7721 in vitro and mouse H=hepatoma carcinoma cell in vivo. METHODS In vitro experiments, MTF assay was used to evaluate proliferational inhibition of P. saponin D on SMMC-7721 cells. In vivo experiments, ICR mice were used to establish H22hepatoma carcinoma cell tumor model. The sup- pression ratio of I-I22 hepatoma carcinoma cell growth was determined. The morphologic and apoptotic body were evaluated by light microscope (HE staining) and electron microscope. RESULTS Medication groups showed dif- ferent levels of SMMC-7721 cells proliferation inhibition after 24 h, 48 h, 72 h with P. saponin D. Marked inhibi- tory effect of Pulsatilla saponin D on the transplanted hepatocellular carcinoma H= was observed in the tumor-bear- ing mice. The inhibitory rates were 38.67% and 41.59% respectively at the dose of 4.5 and 6.0 mg/kg P. sapo- nin D, respectively. (P 〈 0.01 vs. model group). Histopathologicat examination revealed widespread necrosis with massive accumulation of infiltrating lymphocytes and plasmacytes in the tumors. Numerous apoptotic cells and apop- totic bodies were observed in the tumors under the electron microscope. CONCLUSION Pulsatilla saponin D has anti-tumor effects in vivo and in vitro.
