鸟嘌呤核苷酸交换因子(GEF-H1)是一类通过调节Rho GTP酶的GDP/GTP交换,从而调节其活性的一类调节蛋白。GEF-H1作为Rho GTP酶家族的上游信号调节蛋白,通过微管依赖的激活机制以及非微管依赖的激活机制活化,发挥多种生物学功能。GEF-H1作为一种重要的生物学调节因子,在哺乳动物一系列细胞功能中具有重要的作用,如:细胞骨架,细胞屏障功能,细胞周期,细胞分裂,树突棘形态及癌细胞的增殖,凋亡和侵袭转移等。通过对GEF-H1信号转导机制的研究,能够为肿瘤的治疗提供分子靶标。
The Rho guanine nucleotide exchange factor(GEF-H1) can regulate the activity of Rho GTPase through promoting the exchange of GDP for GTP. As a upstream signal-regulated protain, GEF-H1 is activated by microtubule-associated pathway and other ways. Here, we review the roles of GEF-H1 in cell cytoskeleton, epithelial barrier permeability, cell cycle, cell division and dendritic spine morphology. GEF-H1 might also regulate tumor pathogenesis involved in cell's proliferation, apoptosis and migration. GEF-H1 could be a novel therapeutic target in multiple human tumors.
