骨钙素是由骨和牙齿中的成骨细胞特异合成和分泌的一种非胶原蛋白,受多种维生素、激素和细胞因子的调节。骨钙素可维持骨的矿化并作为一种激素调解能量代谢。临床上广泛应用骨钙素诊断和监测骨骼疾病、恶性肿瘤、内分泌疾病及妇科疾病等。研究发现饮食诱导肥胖小鼠连续注射未羧化骨钙素,小鼠体重明显降低。有多种方法可以测定血清骨钙素,其中以放射免疫法应用最普遍。天然骨钙素不易获得,且昂贵,用大肠杆菌原核表达可获得大量骨钙素融合蛋白,有利于骨钙素抗体的开发,为骨钙素生理功能和药理作用的研究奠定了基础。
Osteocalcin which is a non-collagen protein primarily was specific synthetized and secreted by the osteoblast in bone and teeth, and was regulated by many kinds of vitamin, hormone and cytokine. Osteocalcin sustains bone mineralization and behaves as hormone regulating energy metabolism. Detection of osteocalcin has been widely used to diagnose bone diseases, cancer, endocrine diseases and gynecological diseases. In a diet induced obesity model, pellets delivering of recombinant significantly reduced body weight gain, osteocalcin RIA is the most common technical methods of determination of serum osteocalcin in many kinds. Natural osteocalcin are not readily available and very expensive, osteocalcin can be expressed by E. coll. It is helpful to futher study physiological function, pharmacological action and invent osteocalcin antibodies.
