目的:破骨样细胞是指体外诱导培养的破骨细胞,具有破骨细胞的特征,产生的数量较大,基本能够代替原始破骨细胞应用于研究。主要介绍了破骨细胞分化因子、巨噬细胞集落刺激因子、1,25-羟胆钙化醇、白细胞介素6等诱导培养破骨样细胞和其诱导机制的研究进展。资料来源:应用计算机检索Pubmed数据库1990-01/2005-12有关破骨样细胞诱导培养及机制等相关文章,检索词“osteoclasts,ODF,M-CSF,1,25-DihydroxyvitaminD3,IL-6”,限定文章语言种类为English;同时计算机检索中国期刊全文数据库1994-01/2005—04期间的相关文章,检索词“破骨样细胞,破骨细胞”,限定文章语言种类为中文。资料选择:对资料进行初审。纳入标准:①有关破骨样细胞培养方法的研究。②有关破骨样细胞诱导机制的研究。排除标准:重复研究。资料提炼:共收集到119篇文章,排除重复或类似研究,26篇符合要求被选为参考文献。资料综合:①破骨样细胞的诱导培养:破骨样细胞的细胞来源广泛,诱导剂也不同。(砻破骨样细胞的诱导机制:破骨细胞分化因子,巨噬细胞集落刺激因子,1,25-羟胆钙化醇,白细胞介素6等各种细胞因子具有其各自的诱导机制,但相互联系,形成复杂的网络结构,最终通过OPG/RANKL/RANK系统发挥作用。结论:破骨样细胞的诱导培养已逐渐成熟,其具体机制得到初步揭示。破骨细胞分化因子、巨噬细胞集落刺激因子、1,25-羟胆钙化醇、白细胞介素6等各种细胞因子虽然最终通过OPG/RANKL/RANK系统来促进破骨细胞的分化,但各因子间复杂的网络作用仍然需要进一步研究。
OBJECTIVE: Osteoclast-like cells (OLCs) are osteoclasts (OCs) induced in vitro, which own the characteristics of OCs with large quantity, so they can substitute OCs and be used in research. This paper mainly introduces the advances in the methods, identification and mechanisms of OLCs induced by osteoclast differentiation factor (ODF), macrophage colony-stimulating factor (M-CSF),1α, 25-dihydroxyvitamin D3 and interleukin 6 (IL-6). DATA SOURCES: A computer-based online search of Pubmed database was undertaken for articles about the methods and mechanisms of inducing cuhure of osteoclast-like cells published in English between January 1990 and December 2005 with the key word of "osteoclasts, ODF, M-CSF, 1α, 25-Dihydroxyvitamin D3, IL-6". Meanwhile, we searched CNKI for relevant Chinese articles published between January 1994 and April 2005 with the key words of "osteoclast-like cell, osteoclast" in Chinese. STUDY SELECTION: The data were primarily selected. Inclusive criteria: ①research about the methods to culture osteoclast-like cells; ②research on the mechanism of the inducing cuhure of osteoclast-like cells. Exclusive criteria: repeated research. DATA EXTRACTION: Totally 119 articles were received. After excluding the repeated or similar researches, 26 articles were included as references. DATA SYNTHESIS: ①Culture osteoclast-like cell: Osteoclast-like cells have wide resources and different inductors. ②Inducing mechanism of culturing osteoclast-like ceils: ODF, M-CSF, 1α, 25-dihydrixyvitamin D3, IL-6 etc. have their own mechanisms of inducing OLCs, but they contact with each other, and form a complex network, ultimately play the role through OPG/RANKL/RANK system. CONCLUSION: The inducing culture of osteoclast-like cells are gradually progressed, and the mechanisms have also been initially revealed.
