目的:探讨益气温阳护卫法对哮喘大鼠白细胞介素4/信号传导子及转录激活子6(IL-4/STAT6)信号通路的调节作用。方法:50只SPF级雄性SD大鼠随机分为正常对照组、哮喘模型组、益气温阳护卫组、地塞米松组和卡介菌多糖核酸组,每组10只。采用TaqMan Realtime PCR测定哮喘大鼠Th细胞IL-4和STAT6的mRNA表达变化。结果:哮喘模型组IL-4和STAT6的mRNA表达显著高于正常对照组(P〈0.01),益气温阳护卫组、地塞米松组和卡介菌多糖核酸组PBMC中IL-4和STAT6的mRNA表达显著低于哮喘模型组(P〈0.01),且与正常对照组IL-4和STAT6的mRNA表达相当。结论:益气温阳护卫法能够抑制哮喘大鼠IL-4/STAT6信号通路基因的表达,该作用可能是调节Th1/Th2细胞亚群趋于平衡的重要机制。
Objective: To explore the effect of Yiqiwenyanghuwei method on signal pathway of IL-4/STAT6 in asthma rats. Methods: Fifty SPF male SD rats were randomly divided into 5 groups: the normal control group, the asthma group, the Yiqiwenyanghuwei group, the Dexamethasone group, the BCG-PSN group .The mRNA expression of IL-4 and STAT6 in the PBMC of all groups were detected by TaqMan Real time PCR. Results: The mRNA expression of IL-4 and STAT6 in PBMC of asthma group was significantly higher than that of control group(P〈0.01), the mRNA expression of IL-12 and STAT4 in PBMC of Yiqiwenyanghuwei group, the Dexamethasone group and the BCG-PSN group were significantly lower than that of asthma group(P〈0.01), but corresponding with the normal control group. Conclusion: Yiqiwenyanghuwei method can inhibit gene expression of IL-4/STAT6 signaling pathway in asthma rats, the effect may be the important mechanism to regulate Th1 / Th2 cell subsets balance.
