目的:探索建立符合中医寒哮临床发病特点的动物模型。方法:50只清洁级健康雄性SD大鼠随机分为正常对照组(A组)、西医对照组(B组)、中医模型1组(C组)、中医模型2组(D组)和中医模型3组(E组),每组10只。检测各组大鼠肺泡灌洗液(BALF)中细胞变化、BALF中IL4、IFNγ水平变化以及大鼠肺组织病理学的改变。结果:①B、C、D、E组BALF中细胞总数和嗜酸性粒细胞(EOS)百分比均较A组升高(P〈0.01),且C组大鼠BALF中的细胞总数和EOS所占比例明显高于B组(P〈0.01)。②B、C、D、E组大鼠BALF中IFNγ的含量与A组比较均降低(P〈0.01),且C组与B组组间比较存在明显的差异(P〈0.05),B、C、D、E组大鼠BALF中IL4的含量与A组比较均升高(P〈0.01),C、D、E组与B组IL4的含量差异均具有统计学意义(P〈0.05),且以C组的差异最明显(P〈0.01),同时D、E组与C组组间比较具有显著性差异(P〈0.05)。③中医模型组大鼠都表现哮喘炎症反应的病理改变。结论:大鼠BALF中的EOS百分比、IL4和INFγ浓度及病理的改变可能成为中医寒哮动物模型的主要评价指标。
Objective: To explore the establishment of animal model consistent with the traditional Chinese medicine clinical morbility feature of the cold asthma. Methods: 50 Clean healthy male SD rats were randomly divided into 5 groups: the normal control group (Group A), Western medicine control group (Group B), Traditional Chinese Medicine (TCM) mode 1 group (Group C), TCM mode 2 group (Group D) and TCM mode 3 group (Group E), 10 in each group. Test the changes in cells of rats in BALF, the changes in levels of IL-4, IFN-γ in BALF and also the changes in histopathology of rat lung. Results: ①The total number of cells and percentage of EOS in BALF of Group B, C, D, E were higher than that of normal control group (P0.01), and the total number of cells and percentage of EOS in BALF of Group C rats was higher signifi cantly than that of Group B (P0.01). ②The content of IFN-γ in BALF of Group B, C, D, E rats were lower than that of Group A (P0.01),and Group comparison of Group C and Group B is existence signifi cant difference (P0.05). The contents of IL-4 in BALF of Group B, C, D, E rats were higher than that of Group A (P0.01); the content of IL-4 of TCM Model Group C, Group D, Group E comparison with Group B were significant differences statistically (P0.05), and the differences of Group C were the most obvious in of all (P0.01), Chinese medicine model Group D, Group E comparison with Group C were significantly different (P0.05). ③Rats of model group have shown the pathological changes of asthma inflammatory response. Conclusion: Changes of eosinophil percentage, concentration of IL-4 and INF-γ and pathological in BALF of rat may be the main evidence of evaluation to TCM animal model of the cold asthma.
