目的观察消郁颗粒对慢性应激性刺激所致抑郁大鼠的改善作用,明确与皮层和海马神经生长因子(NGF)和脑源性神经营养因子(BDNF)表达的相关性。方法采用慢性应激性刺激制备抑郁大鼠模型,利用蔗糖水消耗、Open-field开场、Morris水迷宫实验观察消郁颗粒对各组大鼠行为学的影响,采用免疫组化法检测额叶皮层和海马区的NGF和BDNF表达。结果与模型组比较,消郁颗粒组体重和糖水消耗体积显著增加,游泳总路程、远站台区路程、近站台区路程、上台潜伏期、近站台区时间和远站台区时间显著缩短,显著增加近/远站台区时间比例(P〈0.05,0.01)。此外,消郁颗粒可显著增加额叶皮层和海马NGF和BDNF的表达,并可呈剂量依赖性(P〈0.05,0.01)。结论消郁颗粒对慢性应激刺激所致抑郁大鼠有一定改善作用,该作用与增加额叶皮层和海马NGF和BDNF的表达有关。
Objective To study the improvement and mechanism of Xiaoyu granules on depressive rats. Methods The behavior of depressive rats,which were formed by chronic stress,were detected by Morris Water Maze,syrup consuming and open-field experiments. Moreover,the expressions of NGF and BDNF in cortex and hippocampus were measured to explain its mechanism. Results Comparing with model group,Xiaoyu granules increased the body weight and syrup consuming obviously,shortened the total swim route,latency,time consuming and route of swimming in distant and nearby area,enhanced time consuming ratio of swimming in nearby and distant area( P〈0. 05,P〈0. 01).Moreover,Xiaoyu granules increased the expressions of NGF and BDNF in cortex and hippocampus markedly( P〈0. 05,P〈0. 01). Conclusions Xiaoyu granules could improve the depressive rats and the mechanism involving with increasing the expressions of NGF and BDNF in cortex and hippocampus.
