目的:研究欣胃颗粒及其拆方对胃癌前病变大鼠Wnt-1及β—catenin的影响。方法:实验分成空白组、模型对照组、胃复春组,益气组、养阴组、化瘀组、解毒组、益气养阴组、益气化瘀组、益气解毒组、养阴化瘀组、养阴解毒组、化瘀解毒组、益气养阴化瘀组、益气养阴解毒组、益气化瘀解毒组、养阴化瘀解毒组、益气养阴化瘀解毒(即欣胃颗粒)组。采用以MNNG为主的四因素联合造模法建立胃癌前病变的大鼠模型,空白对照组、模型对照组给予等量生理盐水灌胃,而其它16组同时给予相应药物灌胃,在16周造模结束后处死大鼠并取材,观察各组大鼠胃粘膜病理改变;采用RealTimePCR技术,检测胃黏膜Wnt信号通路关键因子Wnt-1、β—catenin的基因表达情况。结果:与空白组比较,模型组Wnt-1及β—catenin的基因表达水平明显增高,其差异具有显著性(P〈0.05);中药各组与模型组比较,Wnt-1及β—catenin的表达水平下调,具有统计学意义(P〈0.05)。结论:四法组的干预作用优于三法、两法、及单法组,表明各法之间具有协同增效作用;欣胃颗粒可改善胃癌前病变大鼠胃黏膜病理状况,下调Wnt-1及β-catenin的基因表达水平。
Objective : To study the effect of Xinwei particles and its decomposed recipes on the Wnt - 1 and β- catenin in rats with precancerous lesions of gastric carcinoma. Methods : The rats were divided into normal control group, model group, Weifuchun group, Yiqi group, Yangyin group, Huaxu group, Jiedu group, Yiqi Yangyin group, Yiqi Huaxu group, Yiqi Jiedu group, Yangyin Huaxu group, Yangyin Huaxu group, Huaxu Jiedu group, Yiqi Yangyin Huaxu group, Yiqi Yangyin Jiedu group, Yiqi Huaxu Jiedu group, Yangyin Huaxu Jiedu group and Yiqi Yangyin Huaxu Jiedu group. The rat model with precancerous lesions of gastric carcinoma (PLGC) was established by many pathogenic factors in which MNNG was mainly used. The corresponding drug was given Weifuchun group, Yiqi group, Yangyin group, Huaxu group,Jiedu group, Yiqi Yangyin group, Yiqi Huaxu group, Yiqi Jiedu group, Yangyin Huaxu group, Yangyin Huaxu group, Huaxu Jiedu group, Yiqi Yangyin Huaxu group, Yiqi Yangyin Jiedu group, Yiqi Huaxu Jiedu group, Yangyin Huaxu Jiedu group and Yiqi Yangyin group. Physiological saline was given to normal control group and model group. After 16 weeks,observe the pathological changes in rats' gastric mucosa as well as Wnt- 1 and β-catenin expression. Resuits : The expression of Wnt - 1 and β- catenin in model group was significantly increased compared with those in normal control group. The expressions of Wnt- 1 and β catenin in traditional Chinese medicine groups were reduced compared with those in the model group, and the differences between groups were statistically significant (P 〈 0.05 ). Conclusion : The combination of four therapies for intervention is superior to the three therapies, double therapies and single therapy, and different therap mucosa in rats with PLGC. ies have synergistic effect.
