中医证候的科学本质是中医基础研究中的核心内容,也是揭开中医辨证论治个性化治疗奥秘的关键,但目前对其的认识仍然有限。通过介绍复杂性状遗传学的基本概念,提出将证候作为生命活动表现的复杂性状,运用复杂性状遗传学研究手段,特别是当前迅速发展的全基因组关联分析(Genome-wide association study,GWAS)方法,以解析证候个体化差异的分子遗传基础。除了进行人群的证候GWAS研究,还可利用遗传参照种群小鼠资源进行证候的分子及系统网络GWAS研究。故展望了在中医基础研究中利用模式动物建构合适的中医病-证模型进行GWAS研究的广阔前景,该方法的推广应用将在新的方向上推动中医基础研究的发展。
The scientific nature of TCM Zheng is a treatment based on Zheng differentiation and personalized key for understanding the hallmarks and advantages of TCM in medicine. It is also a fundamental theme of TCM research but remains largely unclear. Here by introducing the concept of complex trait genetics, the authors propose to use related methods, particularly the approach of cutting-edge genome-wide association study (GWAS) to dissect the molecular genetics of the individual differences in TCM Zheng. Moreover, the authors discuss the practicability and necessity to study the molecular and systematic networks underlying Zheng, by taking advantage of genetic reference mouse population resources, and point out the plausible advancement to TCM research and clinic by applying GWAS to validated models of disease-Zheng in the informative mouse population.
