目的研究人bcl—xL基因在显微注射所产生的子代小鼠中的整合、转录和表达情况。方法采用PCR和Southern blot方法检测人bcl—xL基因在小鼠体内的整合;RT—PCR方法检测它们的bcl—xL mRNA水平;用Western blot及免疫组化法检测目的蛋白的表达情况。结果在显微注射所产生的转基因小鼠中,有4只为Southern blot检测阳性,并且在这4只小鼠及其子代中均检测到人bcl—xL的mRNA和过表达的目的蛋白。结论由于在这些小鼠中既有外源基因的整合,又有其mRNA和蛋白质的表达,所以,它们是真正的转基因动物。
Objective To study the integration, transcription and expression of human bcl-xL gene in the transgenic mice generated by microinjection. Methods The integration of human bcl-xL gene in the body of the mice was detected by using PCR and Southern blot. By using RT-PCR and Western blot, the expression of bcl-xL mRNA and protein was assayed respectively in these mice. Results In the transgenic mice after microiniection, there were 4 Southern-blot positive mice, and the expression of bcl-xL mRNA and protein was detected. Conclusion Because these mice not only had the foreign gene integration but also had the foreign mRNA and protein expression, we could conclude that they were true transgenic mice.
