目的研究miR-218在大肠癌与正常组织中表达的差异。方法应用实时荧光定量聚合酶链反应检测60例大肠癌及相应正常组织中miR-218的表达水平,计算其相对表达量。结果60例大肠癌组织中miR-218的相对表达量为0.33±0.12,显著低于对照组,miR-218的表达差异有统计学意义(P〈0.05);miR-218在不同性别、年龄、肿瘤大体形态、组织学类型、临床分期、浸润深度、肿瘤分化程度上表达差异无统计学意义(P〉0.05),有淋巴结转移较无淋巴结转移者表达量低(P〈0.05)。结论 miR-218在大肠癌中呈低表达,其表达水平越低可能提示淋巴结转移越早,miR-218可能作为抑癌基因参与肿瘤的发生、发展。
Objective To study the expression levels of miR-218 in colorectal cancer and normal tis- sues. Methods Using real-time quantitative polymerase chain reaction to detect the expression levels of miR-218 in 60 cases of colorectal cancer and the corresponding normal tissues, and calculate its relative ex- pression. Results The relative expression of miR-218 in the tissues of 60 patients with colorectal cancer was 0.33 + 0. 12, it was significantly lower than the control group, the difference was statistically significant (P 〈0. 05) ;there was no statistically significant difference in the expression of miR-218 in the general form of gender, age, tumor histological type, clinical stage, depth of invasion, and the degree of tumor differentiation ( P 〉 0. 05 ) ; there was lower expression in lymph node metastasis than in those without lymph node metasta- sis (P 〈 0. 05 ). Conclusion miR-218 shows low expression in colorectal cancer, Its lower expression levels may indicate earlier lymph node metastasis, miR-218 may act as a tumor suppressor gene involved in tumor development.
