目的:观察狼疮方含药血清对人近端肾小管上皮细胞(HK-2)过氧化物酶体增殖物激活受体γ(PPARγ)及干扰素(INFγ)和肿瘤坏死因子(TNFα)诱导的白细胞分化抗原40(CD40)、受激活调节的正常T细胞表达和分泌因子(RANTES)表达的影响,探讨该复方体外抗炎效应及其可能机制。方法:制备狼疮方含药大鼠血清;HK-2细胞株体外培养,分组如下:(1)培养基对照组;(2)正常鼠血清对照组;(3)狼疮方含药血清组;(4)IFNγ+TNFα刺激组;(3)刺激+10%狼疮方血清组;(4)刺激+10%正常鼠血清对照组。采用免疫印迹(Western-blot)检测PPARγ蛋白表达,半定量逆转录多聚酶链反应(RT-PCR)检测CD40 mRNA、RANTES mRNA表达水平。结果:(1)正常HK-2细胞基础量表达PPARγ;10%狼疮方含药血清作用下,PPARγ蛋白表达呈现时间依赖性先增高后降低的趋势。(2)正常HK-2细胞基础水平表达CD40、RANTES。IFNγ+TNFα刺激后,CD40和RANTES表达显著升高;10%狼疮方含药血清可显著抑制IFNγ+TNFα诱导的CD40和RANTES表达;正常鼠对照血清对CD40和RANTES表达无明显影响。结论:狼疮方含药血清可能通过激活PPARγ途径抑制人近端肾小管上皮细胞CD40和RANTES表达,从而发挥抗炎效应。
Objective: To observe the effect of Lupus Recipe on the expression of PPARγ and TNFα with IFNγ induced CD40 mRNA and RANTES mRNA on renal tubular epithelial cells (HK-2). Methods:Rats weighted 180- 230 g were randomly divided into control group and treated group. The treated group was treated with Lupus Recipe ( 10 g·kg^-1·day^-1) daily, and the control group was given the same volume of PBS. Rats were sacrificed at 7 days after treatment. Serum of rats were collected. HK-2 cells were cultured in DMEM/F12 with 10% fetal bovine serum in vitro and were divided into following groups: the DMEM medium control group, the normal rat serum group and the Lupus Recipe contained serum group, IFNγ(50 μg·mL^-1 ) + TNFα( 10 ng·mL^-1 ) group, IFNγ(50 μg·mL^-1 ) + TNFα( 10 ng·mL^-1) with 10% drug-contained serum groups and IFNγ(50 μg·mL^-1) + TNFα(10 ng·mL^-1) with 10% normal rat serum group, The expression of PPARγ protein was measured by Western-blotting analysis and the expression of CD40 mRNA and RANTES mRNA was measured by RT-PCR. Results: HK-2 cells expressed low levels of PPARγ,CD40 mRNA and RANTES mRNA. Drug-contained serum of Lupus Recipe can up-regulate the expression of PPARγ protein. Activation of HK-2 cells by TNFα and IFNγ resulted in an enhanced expression of CD40 mRNA and RANTES mRNA; The Drug-contained serum of Lupus Recipe can significantly blocked CD40 mRNA and RANTES mRNA expression in HK- 2 cell line induced by IFNγ plus TNFα. Conclusions:Drug-contained serum of Lupus Recipe up-regulated the expression of PPARγ protein and decreased CD40 and RANTES mRNA expression induced by IFNγ plus TNFα in HK-2 cells. These immune modulation and anti-inflammation effects were partially mediated by PPARγ-dependent pathways and it could be one of the mechanisms of Lupus Recipe in preventing and treating LN.
