本文首先从几种常用的细菌培养基中筛选适合工程菌DH5α/pDH表达的重组人粒细胞-巨噬细胞集落刺激因子(rhGM-CSF)的培养基,其次,对摇瓶培养条件进行了初步优化,着重研究了培养温度、诱导时机,诱导维持时间,接种量对E.coli生长和rhGM-CSF表达的影响。在优化条件下,rhGM-CSF的表达量为26.3%。
Culture medium for recombinant human granulocyte macrophage colony stimulating factor (rhGM-CSF) expressed by engineerng-bactefium DH5α /pDH was selected from several frequently-used bacterium culture medium in this paper. Some shaking-flask fermentation condition was optimized and the effects of culture temperature, time of inducing, expression times on the growth of bacterium and inoculum volume were significantly researched for E.coli growth and rhGM-CSF expression volume. Under the optimal condition, rhGM-CSF expression volume was 26.3%.
