目的通过两种实验方法来确定中药淫羊藿及其单体淫羊藿苷(ICA)在小鼠体内是否具有雌激素样作用。方法将实验动物按体重均衡和随机的原则分为8组:正常对照组、补佳乐组、倍美力组、淫羊藿组和不同剂量的淫羊藿苷组。采用子宫增重实验、ELISA实验法,通过对幼龄小鼠子宫湿重和血清中相关激素的影响,观测是否具有雌激素的作用。结果子宫增重实验中,ICA120 mg组和ICA240 mg组可使动物子宫系数升高明显大于正常对照组,随着ICA剂量的增加,子宫系数在120 mg左右达到一个峰值。在血清雌激素与睾酮含量水平测定实验中可以看出,ICA50 mg组和ICA120 mg组灌药4 d后,能明显升高小鼠血清雌激素水平。并且随着ICA剂量的增加,雌激素水平维持在相对高的一个水平。与正常组相比,ICA50 mg组和ICA120 mg组明显减少血清中睾酮的含量,与正常组差异显著(P〈0.01)。但随着ICA剂量的增加,睾酮的水平逐渐回升,并高出正常对照组水平。结论低剂量ICA可以增加幼龄大鼠子宫重量,并伴随血清中雌激素水平升高和血清睾酮的下降,高剂量ICA并不能继续增加血清中的雌激素含量,但可以持续增加血清睾酮的含量。
Objective To determine whether Herba Epimedii and monosomic icariin(ICA) have estrogenic effects in mice by two kinds of test methods.Methods The experimental mice were divided into 8 groups according to body weight and random principle,including the normal control group,progynova group,premarin group,Herba Epimedii group and ICA groups with different doses.The uterotrophic assay and ELISA were used to observe the estrogen-like effects of Herba Epimedii and ICA on uterine wet weight and serum relevant hormones in young mice.Results In the uterotrophic assay the uterine coefficient increased more in ICA120 mg group or in ICA240 mg group than that in the normal control group,and that reached a peak value with the increase of ICA dose(around 120 mg).In the determination of serum estrogen and testosterone levels after 4-day taking drugs the serum estrogen level increased significantly in ICA50 mg group or ICA120 mg group,and maintained at a relevant high level with the increase of ICA dose.Compared with the normal control group the serum testosterone level decreased in ICA50 mg group or ICA120 mg group with a greatly significant difference(P〈0.01),while when ICA dose increased the serum testosterone level increased gradually and reached a higher level than that in the normal control group.Conclusion A lower dose of ICA can increase the uterus weight of young mouse accompanied by the increase of serum estrogen level and decrease of testosterone level,while higher dose of ICA can not continue to increase serum estrogen level but can in the increase of serum testosterone level.
