目的 探讨8 Hz 130 dB次声作用后谷氨酸转运蛋白2(EAAT2)在脑组织中表达的改变及其意义.方法 SD大鼠40只随机分为对照组及次声作用1,7和14次共4组,次声作用组用8 Hz130 dB的次声接规定次数作用,每次2h.采用Western blot法进行蛋白测定.用实时逆转录酶-聚合酶链式反应法进行mRNA测定.结果 次声作用后EAAT2在蛋白和mRNA水平均呈下调趋势.结论 在次声脑损伤后,谷氨酸在细胞外堆积,可能与EAAT2下调导致谷氨酸重吸收障碍有关.上调EAAT2可能对次声脑损伤后脑组织有保护作用.
Objective The changes of the excitatory amino acid transporter2 (EAAT2) in brain after infrasonic damage in rats and its significance are investigated.Methods Forty SD rats were randomly divided into four groups,namely,control group,1 d,7 d,and 14 d infrasonic damage groups.Rats were exposed to 8 Hz 120 dB infrasound as designed each time for two hours.The Western-blot were used to detect the changes of protein.The mRNA was detected through reverse transcription polymerase chain reaction (RT-PCR).Results Proteins and mRNA of EAAT2 were down-regulated after infrasonic damage.Conclusion Ectocytic glutamate accumulation after infrasound exposure may be related with the disorder of glutamate's reabsorbing.The up-regulation of EAAT2 may protect the brain tissues from the infrasonic damage.
