以小桐子(Jatropha curcas)c DNA为模版,克隆了Jc NF-YB3和Jc NF-YB10基因的CDS序列。序列分析表明该两个基因分别包含549和528 bp完全阅读框(ORF),编码182和175个氨基酸。Blast搜索结果及进化分析结果表明,Jc NF-YB3和Jc NFYB10都与油桐(Vernicia fordii)NF-YB家族基因序列一致性最高且亲缘关系最近。定量分析表明,Jc NF-YB3和Jc NF-YB10在小桐子根、茎、叶、花、果皮和种子中都有表达,Jc NF-YB3在根中表达量最高,Jc NF-YB10在种子中表达量最高。两个基因在种子发育期的表达量变化趋势一致,但是Jc NF-YB10在种子整个生长发育期的表达量都要高于Jc NF-YB3。Jc NFYB3和Jc NF-YB10都在Na Cl、PEG、4℃低温和ABA处理后表达量不同程度上调,推测这两个基因在小桐子中参与抗逆反应机制和植物激素信号传导。
Two genes of NF-YB family(Jc NF-YB3 and Jc NF-YB10) were isolated from Jatropha curcas. The opening reading frame(ORF) of Jc NF-YB3 was 549 bp, encoded 182 amino acids. The opening reading frame(ORF) of Jc NF-YB10 was 528 bp, encoded 175 amino acids. The results of homology analysis showed that both Jc NF-YB3 and Jc NF-YB10 had the highest identity and the closest relationship with NF-YB family of Vernicia fordii. Jc NF-YB3 and Jc NF-YB10 expression significantly in different organisms, Jc NF-YB3 had the highest transcript profile in root, Jc NF-YB10 had the highest transcript profile in seed, the expression level of Jc NFYB10 was a lot higher than that of Jc NF-YB3 during seed growth periods. The expression level of Jc NF-YB3 and Jc NF-YB10 were both induced by Na Cl, PEG, low temperature and abscisic acid(ABA) and those treatments upregulated Jc NF-YB3 and Jc NF-YB10 gene expression. The results suggested that Jc NF-YB3 and Jc NFYB10 both involved in response abiotic stress and signal transduction pathway of plant hormones in J. curcas.
