目的研究7-二氟甲氧基-5、4’-二甲氧基金雀异黄素(7-difluoromethoxy-5,4’-dimethoxy-genistein,DFMG)对溶血性磷脂酰胆碱(lysophosphatidylcholine,LPC)诱导人主动脉内皮细胞(human aorta en-dothelial cells,HAEC)凋亡的影响。方法体外培养HAEC。Annexin V/PI染色流式细胞术(FCM)分析细胞凋亡率。ELISA法测定Caspase-3、8和9活性。Western blotting分析细胞色素c和死亡受体-4、-5表达。结果 LPC(10μmol/L)处理HAEC 24 h,细胞凋亡率为(31.6%±4.1%)(P〈0.001)。DFMG预孵育降低LPC诱导HAEC细胞凋亡率,Caspase-3、-8和-9活化及细胞色素C和死亡受体-5表达(P〈0.05)。结论 DFMG拮抗LPC诱导HAEC细胞凋亡作用与阻断LPC激活死亡受体和线粒体凋亡途径有关。
【Objective】To investigate the effects of 7-difluoromethoxy-5,4'-dimethoxygenistein(DFMG) on human aorta endothelial cell(HAEC) injury induced by lysophosphatidylcholine(LPC).【Methods】HAEC were cultured in vitro.The apoptotic rate was detected by flow cytometry using Annexin V/PI staining.The enzymatic activity of caspase-3,-8 and-9 were determined by ELISA.Cytochrome C and the death re-porter-4(DR4),-5(DR5) expressions of HAEC were determined by Western blot.【Results】Treatment with 10 μmol/L LPC for 24 h,the apoptotic rate of HAEC increased to(31.6 ± 4.1)%(P 0.001).The pretreatment with DFMG significantly decreased induction of apoptotic cell death,the activation of caspase-3,-8 and-9 and the expression of Cytochrome C and DR5 by LPC treatment.【Conclusion】Antagonistic effects of DFMG on HAEC injury induced by LPC is associated with the blockage of LPC-induced death reporter and mito-chondrial pathways.
