为了解波形蛋白在ST细胞中的定位及其与猪瘟病毒(CSFV)衣壳蛋白(C)的共定位关系,本研究采用抗体对波形蛋白的细胞定位进行检测,并将CSFV C蛋白真核表达重组质粒转染于ST细胞中,转染12h和24 h后利用激光共聚焦显微镜观察波形蛋白和CSFV C蛋白在细胞中的定位情况.结果表明,波形蛋白在细胞核周围定位,并且与CSFV C蛋白在细胞中存在共定位现象.本研究为进一步研究波形蛋白在病毒复制过程中的机制奠定基础.
To study the localization of cellular vimentin and classical swine fever virus (CSFV) capsid (C) protein in swine testicle (ST) cells, the recombinant plasmids of pEGFP-CSFV-C expressing the fusion protein of GFP-CSFV-C was transfected into the ST cells and the localizations of CSFV-C and the vimentin were visualized by GFP and antibody against vimentin under Laser confocal microscope at 12 and 24 hours post the transfection, respectively. The results showed that vimentin was localized around the nucleus and also co-localized with CSFV C protein in ST cells, which provided the basis for further studies the function of vimentin in CSFV replication.
