目的:探讨RhoE在胃癌、结肠癌以及相应癌旁组织中的表达情况及临床意义。方法:采用免疫组化的方法检测74例胃癌,32例结肠癌及相应癌旁组织中RhoE的表达水平。结果:RhoE在胃肠道正常组织中普遍表达,在腺体中尤为明显,但在肿瘤组织表达低下或缺失。RhoE在胃癌组织中的染色平均值为1.99±0.28,在相应癌旁组织中为8.64±0.39,明显高于相应肿瘤组织(P〈0.005),并且随着肿瘤分化程度的降低染色强度有下降的趋势(P〈0.005)。RhoE在32例结肠癌组织中的染色平均值为2.45±0.35,在相应癌旁组织中为6.55±0.63,明显高于相应肿瘤组织(P〈0.005),同样有染色强度随着肿瘤分化程度的降低而降低的趋势(P〈0.005)。结论:RhoE在肿瘤组织中表达明显下降,表明它在肿瘤的发生发展中起着重要的负性调控作用,结合最新的研究成果我们推断RhoE很可能是一个重要的抑癌基因。
Objective: To investigate the expression and clinical significance of RhoE in gastrointestinal (GI) carcinoma as well as the corresponding adjacent tissues. Methods: The expression of RhoE was detected by immuno-histochemistry in 74 samples of gastric carcinoma and 32 samples of colon carcinoma, and their corresponding adjacent tissues were taken as control. Results: RhoE was generally expressed in normal gastrointestinal tissues, especially in glands. But its expression level in cancerous tissues was decreased or absent. The stained value for gastric carcinoma was 1.99±0.28, but the adjacent cancerous tissues was 2.45±0.35, significantly higher than that of the adjacent cancerous tissues ( P 〈 0.005). Moreover, there is a declining tendency of the stained value as the poor differentiation of the carcinoma ( P 〈 0. 005 ). The stained value for colon carcinoma was 2.45 ± 0.35, lower than that of the adjacent cancerous tissues (6.55 ± 0.63, P 〈 0.005 ). There was also a declining tendency of the stained value as well as gastric carcinoma( P 〈0. 005 ). Conclusion: The expression of RhoE is significantly decreased in the samples of gastrointestinal carcinoma. So it suggested that RhoE may play a negative role in the generation and development of GI cancer. With the recent research outcomes, RhoE might be an important candidate for tumor supressor gene.
