目的检测成纤维细胞在表皮葡萄球菌刺激下增殖及α一平滑肌肌动蛋白(α-SMA)的表达变化规律,探讨乳房假体植人术后表皮葡萄球菌感染相关包膜挛缩机制。方法MTT法检测普通培养液、不同浓度表皮葡萄球菌SERP62A及SEATl2228裂解液培养的成纤维细胞生长情况;Westernblot及免疫组化法检测不同培养液培养成纤维细胞α—SMA的表达。结果SERP62A对成纤维细胞的生长有明显的促进作用(P〈0.05),SEATl2228对成纤维细胞的生长无明显影响;SERP62A能激活成纤维细胞转化为α—SMA表达阳性的肌成纤维细胞,α-SMA蛋白表达水平明显高于SEATCCl2228组及普通培养液组(P〈O.01),SEATCCl2228组有部分细胞转化为肌成纤维细胞;两组细菌刺激后的细胞第5天及第7天均能维持较为稳定的α—SMA蛋白表达水平;空白组的细胞ccSMA阴性,虽然于培养第5天α-SMA蛋白表达水平有所上升,但第7天时逆转(P〈O.05)。结论SERP62A能促进成纤维细胞增殖,相比SEATCCl2228更有利于成纤维细胞转化为肌成纤维细胞。
OBJECTIVE To investigate the growth tendency and expression of alpha-smooth muscle actin(a-SMA) after the activation of fibroblasts by the Stphylococcus epiderrnidis and discuss the S. epidermidis infection related capsular contracture mechanism after breast prosthesis implantation. METHODS The growth tendency of fibroblast in common culture, SE RP62A and SE AT12228 lysate with different concentrations was tested by the MTT assay. The expressions of alpha-smooth muscle actin(a-SMA)was tested by immunohistochemistry and Western blot in some condition. RESULTS The S. epidermidis RP62A medium could obviously promote the growth of the fibroblasts(P~0.05), however, the S. epidermidis AT12228 medium had no significant effect on the growth of the fibroblasts. The fibroblasts could express more a-SMA when activated by the SE RP62A strains, compared to those troated by SE AT12228 and common culture(P~0.01). Fihroblasts cultured by SE RP62A medium or SE AT12228 medium could express a -SMA steadily on day 5 to day 7. The normal medium fibroblasts started going up with few cell transition into myo-fibroblast, then went doum typifed as myo-fibroblast on day 7(P~0.05). CONCLUSION SE RP62A could improve the multiplication of fibroblasts. Compared with SE ATCC12228, SE RP62A could help fibroblasts transfer to myofibroblast better.
