目的:研究蜘蛛香环烯醚萜类成分ethoxyviburtinal-11对大鼠结肠纵行肌细胞的直接作用及初步机制探讨。方法:原代培养大鼠结肠纵行肌细胞,观察不同时间、不同浓度的ethoxyviburtinal-11对大鼠结肠纵行肌细胞的作用及ethoxyviburtinal-11对工具药5-羟色胺(5-HT)、乙酰胆碱(Ach)和去甲肾上腺素(NE)引起结肠纵行肌细胞收缩效应的影响。结果:当浓度为4.04mmol/L,作用30s时,ethoxyviburtinal-11对结肠纵行肌细胞具有最强的舒张效应。ethoxyviburtinal-11对5-HT引起的结肠纵行肌细胞收缩效应有显著抑制作用,而对Ach、NE的效应无显著性影响。结论:ethoxyviburtinal-11对结肠纵行肌细胞具有直接的舒张作用,其舒张作用可能与5-HT有关。
Objective: To observe the effects of ethoxyviburtinal-11, a constituent of valeriana jatamansi jones, on rats' colon longitudinal muscle cells and its mechanism. Methods: The primary cultured rat colon longitudinal muscle cells were administrated by ethoxyviburtinal-11 at different time and concentration. The shrinkage effect induced by tool medicines including serotonin(5-HT), acetylcholine(Ach) and norepinephrine(NE) was observed after the administration of ethoxyviburtinal-11. Results: Ethoxyviburtinal-11 had a strongest relaxed effect on primary cultured rat colon longitudinal muscle cells when the concentration was 4.04mmol/L and the time was 30 s. Ethoxyviburtinal-11 could significantly inhibit the shrinkage effect induced by 5-HT but Ach and NE. Conclusion: The ethoxyviburtinal-11 could directly make the rats' colon longitudinal muscle cells relax, which may be related to 5-HT.
