为了探究左旋咪唑(LMS)作为免疫增强剂对金黄色葡萄球菌粘附素 ClfA 重组 DNA 疫苗免疫效果的影响.分别用含左旋咪唑的 ClfA 重组 DNA 疫苗、ClfA 重组 DNA 疫苗及空载体免疫昆明白小鼠,通过巨噬细胞吞噬试验、MTT 实验、免疫血清对金黄色葡萄球菌识别能力、酶联免疫吸附试验和攻毒试验验证免疫效果.结果表明,含左旋咪唑 DNA 疫苗组在巨噬细胞吞噬指数、脾脏淋巴细胞增殖、免疫血清对金黄色葡萄球菌识别能力方面有增强作用,与其他两组相比差异显著(P <0.05),含左旋咪唑 DNA 疫苗组小鼠抗体水平及攻毒实验保护率高于单独 ClfA 重组 DNA 组和空载对照组,LMS 能够对 ClfA 重组 DNA 疫苗免疫效果起到增强作用.
In oder to explore the effects of the levamisole (LMS)as immune enhancing agent on the im-mune effect of DNA vaccine based on the Staphylococcus aureus adhesin recombinant ClfA,Kunming mice were immunized with ClfA DNA vaccine plus LMS,ClfA DNA vaccine alone,and empty vector,respec-tively.The immune effects were verified by the following:the test of macrophage swallowing,MTT test, the distinguishing capacity of Staphylococcus aureus ,test of enzyme linking immunosorbent absorption and challege test.The results showed that compared with other two groups,the phagocytic index of mouse’s macrophage,the T lymphocyte proliferation response of spleen,Staphylococcus aureus recogni-tion ability of antibody induced by ClfA DNA vaccine can be significantly enhanced by LMS (P < 0.05). Furthermore,the protective response and the antibody level of mice immunized with ClfA DNA vaccine co-administrated with LMS were higer than those of immunized with DNA vaccine and empty vector alone. These results obviously indicated that LMS has good enhance effect on the immunogenicity of ClfA DNA vaccine.
