目的探讨线粒体DNA 5178A/C多态性与2型糖尿病肾病(DN)的相关性。方法将1039例2型糖尿病患者分为无糖尿病肾病(DN-0)组和糖尿病肾病(DN)组,后者进一步分为微量蛋白尿(DN-1)组和大量蛋白尿(DN-2)组,进行病例对照研究。采用聚合酶链反应-限制性片段(PCR-RFLP)技术检测线粒体DNA5178A/C多态。结果(1)2型糖尿病者尿白蛋白肌酐比值(ACR)与5178A/C多态无相关。(2)DN-0,DN-1及DN-2这3组间基因型频率的差异无显著性。结论线粒体DNA 5178A/C多态性与中国2型糖尿病患者ACR水平无相关,与糖尿病肾病无相关。
Purpose To observe the relationship between diabetic nephropathy(DN) and the mitochondrial DNA 5178 adenine/cytosine (mt 5178 A/C) polymorphism in Chinese type 2 diabetic patients. Methods A case-control study for a total of 1039 Chinese type 2 diabetic patients including 524 patients without diabetic nephropathy and 515 patients with diabetic nephropathy for controls was performed. The patients with diabetic nephropathy (DN; n = 515)were further subdivided into microalbuminuric patients (DN1 ; n = 430) and macroalbuminuric patients (DN2; n = 85). The mt 5178 A/C was genotyped by the polymerase chain reaction-restriction fragment length polymorphism method (PCR-RFLP). Results (1) The Mt 5178 C genotype was not associated with the albumin-to-creatinine ratio (ACR). (2) No significant difference in genotype distribution was observed among the DN0, DN1 and DN2groups. Conclusions The Mt 5178 C/A polymorphism was not associated with the ACR and DN among Chinese type 2 diabetic patients.
