克隆了大黄鱼小G蛋白家族中的Rab11基因,测序查明其c DNA序列全长1373 bp,其中5’非编码区为129 bp,3’非编码区为587 bp,开放阅读框为657 bp,编码218个氨基酸;其氨基酸序列与罗非鱼、斑马鱼等鱼类Rab11蛋白序列同源性在95%左右,与人类、大熊猫等物种的同源性也达到90%以上。Rab11基因在大黄鱼脾脏、鳃、肾脏、皮肤、肝脏、血液、肠、心脏和胃9个组织中均有表达,在血液、肝脏中表达量最高,在胃中表达量最低;溶藻弧菌刺激后大黄鱼肝脏、肾脏和脾脏组织中Rab11基因的表达量均明显上调,提示Rab11在大黄鱼的抗病免疫反应中有着重要的作用。
Large yellow croaker( Larimichthys crocea) is one of the most important and largest yield mari-culture fish species in China. However,in recent years,infectious diseases caused by various pathogens are more and more severe. There is an urgent need to understand the mechanism of innate immune in large yellow croaker to control the diseases and for long-term sustainability. In this investigation,a Rab11( LycRab11) gene was obtained from the large yellow croaker. The full length of Rab11 c DNA was 1373 bp,containing a 129 bp 5'-untranslated region( UTR),a 587 bp 3'-UTR and a 657 bp open reading frame( ORF) which encoded 218 amino acids. The sequence of large yellow croaker Rab11 protein shared up to95% identities with those of many fish,such as tilapia and zebrafish,and 90% identities with those of human and panda. Quantitative real-time RT-PCR analysis revealed the highest expression of LycRab11 gene in blood and liver and the weakest expression in stomach. Time couse analysis revealed that LycRab11 expression was obviously up-regulated( after 12 h) in liver,kidney and spleen after immunization with Vibrio parahaemolyticus( P 0. 05). These results suggested that Rab11 protein might play an important role in the immune response in large yellow croaker and this will establish the foundation for the further study of character and function of the Rab11 gene in the large yellow croaker.
