目的：观察中药双黄补对人牙龈成纤维细胞由机械划痕损伤、内毒素脂多糖（LPS）损伤所致的保护作用。方法：采用组织块法体外培养人牙龈成纤维细胞。水提醇沉法制备双黄补提取液。采用四甲基偶氮唑蓝法测定培养细胞增殖活性的变化。建立体外细胞划痕创伤模型,加入双黄补培养24 h观察细胞迁移。绘制生长曲线观察双黄补对LPS介导的成纤维细胞增殖能力的影响。结果：与对照组相比,中药双黄补作用于人牙龈细胞后,能明显促进细胞的增殖,加快细胞的移动速度,改善由LPS介导的细胞损伤后的增殖能力。结论：中药双黄补能促进人牙龈成纤维细胞的增殖,对人牙龈成纤维细胞由机械划痕损伤、LPS损伤具有较好的保护作用。

OBJECTIVE To observe the protective effect of Chinese medicine Shuanghuangbu on human gingival fibroblasts（HGFs） injury induced by mechanical and LPS injury.METHODS HGFs were cultured in vitro by using the method of tissue piece.Shuanghuangbu extract was prepared with water extraction and alcohol precipitation method.Proliferation activity of HGFs was determined by MTT.The in vitro cell scratch wound model was set up to observe the effect of Shuanghuangbu on the migration of HGFs after 24 hours of culture.By drawing the growth curve of HGFs,the effect of Shuanghuangbu on proliferation was observed in the cultured-cells acted by LPS in vitro.RESULTS Compared with control group,Shuanghuangbu could reinforce proliferation activity of HGFs cultured in vitro,accelerated the migration of HGFs and improved the proliferation activity of HGFs after injury induced by LPS.CONCLUSION Shuanghuangbu can reinforce proliferation activity of HGFs cultured in vitro and has the protective effect on mechanical injury and LPS injury.
